Polanski, who pleaded guilty to unlawful sex with a 13-year-old girl in 1977 before fleeing the US, was arrested in Switzerland last week. He is now facing extradition.
Several celebrities, including Woody Allen, Penelope Cruz and Whoopi Goldberg, have supported the director and asked for his release.
However, writing on her Twitter page, Alley said that he should be treated the same as anyone else.
"JUST FOR THE RECORD... RAPE IS RAPE," she wrote. "This is one Hollywood star who does not CELEBRATE or DEFEND Roman Polanski."
She criticised Goldberg's initial remarks that it hadn't been "rape-rape" and added: "I just can't believe twisting it into anything else... every criminal JUSTIFIES his or her actions... but do WE have to also?
"I don't seek PUNISHMENT for people, including Polanski. I seek accountability, justice and rehabilitation. In that order.
"If I CAUGHT someone raping MY daughter... 'JUSTICE' would come in the form of a 45."
