The Jolie-Pitt Foundation has donated the money to Doctors Without Borders/Médecins Sans Frontiéres following the 7.0-magnitude quake that hit the Caribbean island on Tuesday afternoon.
"It is incredibly horrible to see a catastrophe of this size hit a people who have been suffering from extreme poverty, violence and unrest for so many decades," said Jolie.
Pitt added: "We understand the first response is critical to serve the immediate needs of countless people who are now displaced from their homes, are suffering trauma, and most require urgent care."
The couple previously toured Haiti in 2006 and attended a party to celebrate the one-year anniversary of Wyclef Jean's Yele Haiti charity.
Other stars who have called for funds to help the survivors include House co-stars Olivia Wilde and Omar Epps, Heroes stars Jimmy Jean-Louis and Greg Grunberg and Wyclef Jean.
