The TV host was performing at the Channel 4 Comedy Gala when he asked the 20,000-strong crowd to scream the c-word.
Ross stated that he wanted to "break the record for the most people saying c**t at the same time". When the audience obliged he joked: "I'm used to having that shouted at me."
The show, recorded at London's O2 arena, features 25 comics including Michael McIntyre, Sean Lock and Alan Carr. The event will be broadcast on Monday, April 5 on Channel 4 at 9pm.
