According to People, the actor - who plays Dean Winchester in the CW drama - tied the knot with Harris (Rachel Gatina) at a ceremony in Dallas on Saturday.
The couple have been dating for three years and confirmed their engagement last November. A guest at the nuptials is quoted as saying that the pair "looked happier than ever".
Ackles's Supernatural co-stars Jared Padalecki (Sam Winchester) and Genevieve Cortese (Ruby) married in February.
He recently wrapped filming on the fifth season of Supernatural, while Harris can currently be seen alongside Jennifer Lopez in The Back-Up Plan.
Ackles and Harris have yet to comment.
