Two claims of sexual harassment have been made against the star this month relating to his work on documentary I'm Still Here, which follows Joaquin Phoenix's rap career.
Cinematographer Magdalena Gorka filed a $2.25m (£1.43m) lawsuit against Affleck on Friday.
Gorka also claims that she was subjected to a hostile work environment.
The Gone Baby Gone star's representative told TMZ: "[It's] absolute fiction. [It's] fishy… [Gorka] hasn't worked for the movie since April 2009… it's interesting that she hasn't filed until now."
She added: "Several crew members have told us she was inappropriate on set."
A week before Gorka's lawsuit Affleck was sued for $2m (£1.2m) by producer Amanda White.
