According to the AP, the live stream from the New York show will be webcast on YouTube later this month.
The webcast will offer a number of interactive elements, including the ability to switch camera shots.
"It's not necessarily a Spike Lee joint, it's a John Legend and the Roots joint," Lee joked about the event.
Lee's recent films include the documentaries If God is Willing and da Creek Don't Rise, Passing Strange and Kobe Doin' Work.
"I like doing stuff live. Especially with someone like Kobe. I can't stop the game. You got to be ready for almost anything. We can't stop this concert, either. Going to have to go with the flow," the director said.
The concert is part of a series presented by American Express. The first in the series was directed by Terry Gilliam and featured Arcade Fire performing in Madison Square Garden.
