The American comedian, who judges NBC's Last Comic Standing, is in a critical but stable condition at the Robert Wood Johnson University Hospital in New Brunswick, New Jersey, according to the New York Post.
The 44-year-old apparently collapsed on Saturday night in his hotel room, but the incident is not believed to have been a suicide attempt.
A source at Giraldo's management company The Collective confirmed: "It was an accidental overdose, and he is in critical condition."
The comic has recorded two half-hour specials for Comedy Central, and was one of the team captains in a pilot episode of an American version of Have I Got News For You last year.
