The two actors will apparently alternate between the two lead roles - Dr Frankenstein and the monster - on a nightly basis.
Boyle - who is to direct the production - told the Daily Mail that the constant role changes will keep the show fresh, confirming: "They'll share it. One night one will be Frankenstein's [monster], while the other will play his creator Victor."
Cumberbatch - who is best known for playing the eponymous detective in BBC's Sherlock - earned an Olivier Award nomination for his performance in a 2005 production of Henrik Ibsen's Hedda Gabler.
Miller - the ex-husband of Angelina Jolie - has previously worked with Boyle on Trainspotting, in which he played conman Simon 'Sick Boy' Williamson.
The production marks Boyle's first stage show in 15 years, and is due to open at the National Theatre in February 2011.
