The comedian took to his Twitter page on Thursday to confirm that he plans to star in a new production of writer-director Neil LaBute's Fat Pig.
He wrote: "I promised unexpected twists & turns right? I'm making my debut on Broadway this spring In the Neil LaBute play Fat Pig. Beyond excited. (sic)"
According to CNN's Marquee Blog, Cook is to play a man who torments his best friend for falling in love with an overweight woman.
The staging will also mark LaBute's first Broadway production, although he has directed several critically acclaimed films including 1997's Your Friends & Neighbors and the 2010 comedy Death At A Funeral.
Fat Pig received an Olivier Award nomination for 'Best New Comedy' during its London run in 2008.
