The actress fought off competition from Dame Maggie Smith in second place and Mark Rylance in third.
Dench, who won her first Olivier Award in 1977 for Macbeth, was thrilled by the news.
"I'm completely at a loss for words, but it's one hell of a thing to live up to," she said.
Lord of the Rings star Ian McKellen came in fourth, Sir Laurence Olivier was fifth, followed by Paul Scofield, Sir John Gielgud, Sir Michael Gambon, Vanessa Redgrave and Sir Ralph Richardson.
