The actress will star alongside Dane Cook and Josh Hamilton in the production, reports The AP.
Neil LaBute is directing the play, which opens at New York City's Belasco Theatre on April 26.
The play is billed as a "darkly comic" story that centres on a man named Tom, played by Hamilton, who falls in love with a plus-sized young woman named Helen.
Stiles will play Jeannie, Tom's ex-girlfriend.
The 29-year-old actress recently wrapped a stint on Showtime's Dexter. She has previously starred in the Broadway revival of David Mamet's Oleanna and a production of Shakespeare's Twelfth Night in Central Park.
LaBute, the film director behind Nurse Betty and In the Company of Men, is making his Broadway debut with Fat Pig.
