Galliano was suspended from his role at Christian Dior last week after allegedly using racial slurs against a couple while drinking at a bar in Paris.
A video from a separate incident later surfaced in which the designer appeared to make further offensive comments.
In a statement, Portman - who was named 'Best Actress' at the Academy Awards on Sunday for her role in Black Swan - said that she was "deeply shocked and disgusted" by the contents of the tape.
"In light of this video, and as an individual who is proud to be Jewish, I will not be associated with Mr Galliano in any way," she added. "I hope at the very least, these terrible comments remind us to reflect and act upon combating these still-existing prejudices that are the opposite of all that is beautiful."
Portman, who is currently expecting her first child, is the new face of Christian Dior's Miss Dior Cherie fragrance.
Galliano has denied making anti-semitic remarks.
