The Late Night host has filmed a series of linking narrations, which will be shown on monitors to those taking the famed Studio Tour beginning this spring.
"We really had fun with filming my role," Fallon said in an official statement.
He continued: "The tour itself is a classic, a really great experience. For anyone with any interest in show business, it's all there: the glamour, the glitz, all the hard work that goes into making a movie or a TV show can be experienced on the Universal Studios Hollywood Studio Tour. I'm very happy to be a part of it."
The Studio Tour was opened in 1915 and has since featured exhibits from popular films including Psycho, Back To The Future and Jurassic Park.
Along with Fallon's new narration, the tour has added the King Kong 360 3-D attraction, which was created by acclaimed director Peter Jackson and opened last summer.
In March, Fallon said that he's proud to be celebrating his second anniversary as the host of Late Night.
