The Australian comedian and TV star, who is best known for appearing on current affairs series The 7pm Project and Rove McManus's former chatshow, welcomed daughter Sadie May at 10.25am on Saturday.
According to The Daily Telegraph, Hughes's wife Holly and baby girl are both doing well in a Melbourne hospital.
The 40-year-old thanked fans for their support on Twitter, writing: "She was delivered the old-fashioned way, mum light on the drugs, heavy on the screaming."
The couple already have a son called Rafferty, who Hughes joked now calls his little sister "the Wah Wah".
Hughes took part in the Australian electoral campaign last year, along with other stars such as fashion designer Ruby Rose, Neighbours actor Matthew Werkmeister and model Tahnee Atkinson.
At the time, he said: "Nothing gets me going more than people whinging about governments or politicians when they won't even vote. If you want to have a say, do something about it and get on the roll because your vote is worth just as much as… Nicole Kidman's."
