The guitarist, who established the prog-rock group in 1965 with Roger Waters, Nick Mason and Richard Wright, died in 2006 at the age of 60, leaving behind a collection of artwork, much of which was later auctioned.
According to The Mirror, a one-off A4 print owned by Barrett's former girlfriend Libby Gaudsen was taken from the wall of Idea Generation Gallery on Saturday (April 9).
The self-portrait was produced in the early 1960s, prior to Barrett's rise to fame and well-publicised drug use.
Gaudsen said of the incident: "I am very upset at the theft of the painting. It has huge personal value to me and I am appealing for its safe return."
According to management at the East London gallery, the painting was taken from the Chance Street site between 2.15pm and 3pm.
A reward of £2,000 has been offered for safe return of the portrait.
