The Celebrity Apprentice star has been forced to defend accusations of racism, after he publicly questioned the president's US citizenship and launched an investigation into Obama's background.
During a conversation about Trump and his recent comments on Thursday's edition of the Late Show, Letterman was asked by guest Dr Phil whether he believed the mogul's suspicions of Obama were racially motivated.
"Well, yes," he replied. "If he comes back on this show, and I am not sure we want him back on this show under these circumstances, he ought to be prepared to apologise just for that kind of behaviour."
Trump, who previously argued that the US is "going to hell" under Obama, has expressed interest in running for office in 2012.
Stars including Cher, Robert De Niro and Chris Rock have spoken out against Trump's attack on Obama and his presidential ambitions.
