The How I Met Your Mother star was confirmed to helm the event last week after previously presiding over the 2009 telecast.
"I'm honoured and stoked to have been asked to host the 2011 Tony Awards," he said in a press release. "It should be a great show - we've got a fantastic mix of live performances [and] a few secret surprises."
He went on to joke: "Since they're closed for retooling, we're using all the rigging equipment from Spider-Man [Turn Off the Dark]. What could possibly go wrong?"
Full nominations for the Tonys were announced last week, with South Park creators Matt Stone and Trey Parker leading the nominations for their musical The Book of Mormon.
The 65th annual Tony Awards will air June 12 on CBS and will be broadcast live from the Beacon Theatre in New York.
