The Social Network actor, who wrote the script, will also star in the production, which is scheduled to debut at New York's legendary Cherry Lane Theatre beginning in October, reveals The Hollywood Reporter.
The story focuses on "two friends who view themselves as open-minded liberals - one blogs about American imperialism, the other is finishing a PhD in Black Studies - and how those self-perceptions are challenged when a young Filipina woman becomes their new roommate", according to the website.
Asuncion will be directed by Kip Fagan and will start previews on October 12 with an official opening following on October 27.
Eisenberg was recently rumoured to be in talks to join Squid and the Whale director Noah Baumbach's new project While We Were Young.
He recently signed up for a role in the upcoming dark comedy Predisposed alongside Melissa Leo.
