The Event star will play Stanley Kowalksi, a rising member of the industrial class, according to Entertainment Weekly.
Marlon Brando and Alec Baldwin previously played the role.
Underwood can be seen next in The Art of Getting By opposite Emma Roberts and Freddie Highmore.
The 46-year-old played President Elias Martinez on the recently-cancelled NBC series The Event. It was rumoured that the show might make the move to SyFy, but the network denied those reports earlier this month.
A Streetcar Named Desire is expected to open in the spring of 2012.
