In late 2010, the Lopez Tonight host commented that he was interested in seeking LA's top office, but his rep later suggested he was merely joking.
Lopez has now revealed that he is seriously mulling a mayoral campaign in "eight years" to inspire Los Angeles residents to take pride in their city.
"This is a great city, and it's my city. I believe that there are certain things that could be taken care of that you don't need a strong political background in," the comic told CNN's Piers Morgan.
He continued: "Architecture downtown is beautiful and people live here, but they don't have a pride in Los Angeles that they did years ago, when I was growing up."
Lopez also threatened to leave the US if rumoured presidential candidate Sarah Palin defeats Barack Obama in the 2012 election.
"If Sarah Palin becomes president at any point, I will say allegedly, I will move to Canada. I will go further North. I've been South, I will go further North," the star joked.
Lopez recently finalised his divorce from Ann after 18 years of marriage.
Piers Morgan Tonight airs weeknights at 9pm ET on CNN and at 8pm UK time on CNN.
