The Blue Valentine actress admitted that she still feels the impact of the Dark Knight star's death three years on.
"It's changed how I see the world and how I interact on a daily basis," Williams told Vogue. "It's changed the parent I am. It's changed the friend I am. It's changed the kind of work that I really want to do. It's become the lens through which I see life - that it's all impermanent."
The 31-year-old went on to say that she is open to another romance in the future.
"Relationships have always seemed mysterious, and therefore worth exploring," Williams said. "I'm single, so it's still kind of a mystery - a worthwhile mystery, one that I want to be on the scent of."
She concluded: "I'm not lonely, and I think that has a lot to do with what's on my bedside table rather than what's in my bed."
Williams previously described the time after Ledger's death as "a year of very magical thinking", saying that in some ways she misses that special time.
The actress, who gave birth to a daughter named Matilda with Ledger in 2005, said in December that the couple's child will watch the actor's film Brokeback Mountain when she gets older.
