Tahrir Square was the site of many rallies that helped topple the regime of former Egyptian President Hosni Mubarak in February.
Demonstrators have gathered again in the public square in the last few days to urge Egypt's military government to halt the same emergency laws that Mubarak allegedly used to quell unrest among his people.
Penn waved an Egyptian flag and called for protestors at the 'Reclaiming the Revolution' rally to continue fighting for freedom, reports newspaper Al-Ahram.
"The world is inspired by the call for freedom by the courageous revolution of Egypt for its freedom," Penn said. "Clearly that is not a completion overnight, there are still struggles forward, there are constitutional issues, there is... a transition of power from the military to the people."
Penn appeared at the rally alongside Egyptian actor Khaled el-Nabawi, who said protestors' ultimate goal is to ensure "that Egypt is safe".
Parliamentary elections in Egypt are to be held in November.
