The horse collapsed and died on 54th Street on Sunday just before it was to begin a full day of carriage rides around the city.
Glee star Michele wrote on Twitter that she was heartbroken to see the animal die and called for an end to carriage rides.
She tweeted: "Just woke up to the saddest news about one of the carriage horses dying the midtown NYC. This has to stop! Please :("
Michele and PETA launched a campaign to end horse-drawn carriage rides earlier this year.
The actress claimed that the horses are "deprived of everything that makes their lives worth living" and asked her fans not to ride on the carriages.
Earlier this week, Michele revealed that she was grossed out by one of Glee's upcoming sex scenes.
Glee returns on Tuesday, November 1 at 8/7c on Fox. The series airs on Sky1 and Sky1 HD in the UK.
