Simpson was acquitted of the double murder of his ex-wife and her friend Ron Goldman in 1995, but was later found guilty of the pair's wrongful deaths in a civil proceeding.
The former pro footballer, who is currently in prison on armed robbery and kidnapping charges, has always maintained his innocence.
Jenner has now revealed in her upcoming memoir that she feels guilty for snubbing a meeting with Brown Simpson on the morning of her murder, in which her friend was about to confide details of "17 years of abuse" and ask her to be "a witness".
"I instinctively knew that in some way OJ had something to do with her death," she writes in Kris Jenner...And All Things Kardashian. "I truly couldn't believe she had been so betrayed by the person who she had once loved most.
"That OJ would be so destructive and selfish and jealous that he would do that to her was just mind-blowing to me. All these thoughts were running through my mind. This can't be true."
On the discovery of Brown Simpson's death, she recalls screaming: "'Oh my God! It's too late! It's too late!'... The realisation that she had wanted to confide in me hit me so hard... And I had let her down."
Jenner's lawyer ex-husband Robert Kardashian played a key role in securing a not guilty verdict for Simpson.
In June, Oprah Winfrey denied rumours that Simpson had confessed to the murders in a conversation with her producers.
