The Hollywood actor will star in a new production of The Sunshine Boys alongside Richard Griffiths. The play by Neil Simon follows two veterans who return for one last performance together.
It was first staged on Broadway in 1972 before being made into a film starring Walter Matthau and George Burns, with Burns receiving an Oscar for his performance.
The play will run from April 27 to July 28 at the Savoy Theatre, and will be directed by Thea Sharrock.
DeVito starred in the original Broadway production of One Flew Over the Cuckoo's Nest in 1971, while Griffiths has won Olivier and Tony awards for his role in Alan Bennett's play The History Boys.
Danny DeVito was awarded a Hollywood Walk of Fame star in August last year.
