The comedian announced during his recent CrazyStupidPolitics webcast that he will give the sizeable donation to Priorities USA Action, a super political action committee promoting Obama's election.
"This is the wisest investment I think I could make," Maher said of his donation.
DreamWorks Animation's Jeffrey Katzenberg is among the other primary supporters of Priorities USA Action.
The group is not officially associated with the Obama campaign, but represents a variety of private supporters of the president.
Priorities USA Action executive Bill Burton said in a statement: "We appreciate Mr Maher and others have so generously stepped up to the plate to fight back."
Maher notably supported Obama's 2008 presidential campaign as well as Senator John Kerry in his 2004 campaign against George W Bush.
