Ed Byrne, Rory Bremner and Mitch Winehouse are among the stars who will perform on Monday, May 14 at the Prince Edward Theatre in Soho for the charity.
Byrne said: "I find it hard to believe that in a city like London, there are still so many young homeless people struggling to make something of their futures.
"This performance is a great chance to see some of Britain's favourite comics share the stage with young talent."
He added: "I urge everyone to buy a ticket and have a laugh while helping to support those who need it most."
