She will be joined by Loose Women's Lisa Maxwell, who recently asked Middleton to take part after meeting at a party.
"I work with Centre Point and William's our patron. We had a little do and they both came down," Maxwell told The Mirror.
"She asked me about sleeping out, so I said, 'Next time I do it, you're doing it with me. No excuses, he's done it'. She said, 'You're on'."
Prince William had previously spent a night living on the streets in 2009 for the charity, being joined by Centre Point's chief executive Seyi Obakin near Blackfriars Bridge.
Princess Diana was also a patron of the charity, which aims to improve the lives of homeless people throughout the UK.
