Paltrow and Joel Gallen are producing the third cancer fundraiser for a September 7 airing on the broadcast networks ABC, CBS, Fox and NBC.
HBO, VH1, the Lifetime Movie Network, the MLB Network, Logo, mun2, Palladia and Bio will also simulcast the special event.
The telethon will feature musical performances and testimonials from cancer survivors as well as a celebrity phone bank.
Paltrow said of the third Stand Up to Cancer telethon: "This broadcast has become a global call-to-action for all those touched by cancer. Like so many people, I know what it's like to lose a family member to this disease, and I'm honoured to stand up in my father's memory."
The Iron Man 3 actress's father Bruce Paltrow died from complications from oral cancer in 2002.
Stand Up to Cancer most recently aired in 2010 and featured appearances by Ryan Seacrest, Lance Armstrong, Diane Lane and George Clooney.
