The comic has signed a two-book deal with publisher HarperCollins, with the first scheduled to hit shelves next autumn.
The firm described Walliams' debut as "a humourous, classic tale that features an engaging boy hero". It added that the book would be aimed at children aged 8-12, but would also be enjoyable for older readers.
Walliams said: "I am thrilled to be writing a children's book, and hope the story will be funny and thought-provoking."
Ricky Gervais scored a bestseller in 2004 with his Flanimals series, while Madonna, Jordan, comedian Steve Martin and the Duchess of York have also written children's books.
Geri Halliwell signed a deal in April to pen six books about nine-year-old Ugenia Lavender.
Walliams is to play Frankie Howerd in a BBC Four biopic to air next spring.
