Publisher HarperCollins described the children's novel as possessing "sharp humour and vivid characters" and telling a story "about what happens when an ordinary boy does something extraordinary and the way that people, even the petty and cruel, can surprise you in the end".
Speaking to the Press Association, Walliams said: "I am thrilled beyond measure to be working with the legendary Quentin Blake.
"I have adored his work since I was a child, and having him illustrate the book is a great honour."
Blake is famous for illustrating the books of children's author Roald Dahl with a distinct pen and ink and watercolour style.
The Boy in the Dress is due to be released in October.
