The 82-year-old magazine chief claimed that the teenage star would be "welcomed" by the adult publication when she reaches the legal age.
Cyrus caused controversy last month when she appeared topless, covered only by a blanket, on the cover of Vanity Fair.
The actress released an apology to her fans after the photographs were released, while a representative for the 15-year-old claimed she had been manipulated by the magazine.
Commenting on the pictures, Hefner said: "I think to make such a big to-do over something as innocent as those photos, I think is a reflection on how schizophrenic America is about sexuality."
