BBC Radio 1 DJ Moyles has been cast as King Herod in the play, while Mel C will portray Mary Magdalene. Tim Minchin was also cast as Judas.
The Spice Girls singer revealed that she has been a big fan of the Lord Andrew Lloyd Webber musical from a young age, and is looking forward to embarking on a UK arena tour revival.
"Musical theatre has been a love of mine for a very long time. The 1980s was a time where Andrew Lloyd Webber became the king of musical theatre. But Jesus Christ Superstar has been with me all of my life," she told Daybreak.
On Moyles's casting, she said: "If people know the show they'll understand the casting, King Herod is a fun role. I've heard Chris sign before, he can sing, he's got a voice."
Mel C has also been taking part as a judge with Jason Donovan during the callback stage of the Superstar search, which is looking for a new singer to play Jesus in an upcoming ITV1 show.
She said: "Jason Donovan has been a dream, we've had lots of fun. The boys have been incredible."
The singer also gave an update for the upcoming Spice Girls musical Viva Forever, which was recently rumoured to have been delayed due to her alleged rift with Mel B.
She added: "We're getting very close to getting the show on the theatre. It should show at the end of the year, if not early the next."
The Jesus Christ Superstar tour is set to kick off on September 21, with tickets going on sale this Friday (May 18).
