Winfrey's series comes to an end today, more than 25 years after it debuted as a local morning show on Chicago television.
Rutherford - a self-professed admirer of the host - predicted that Winfrey's dedication to her audience will ensure that she'll one day return to the talkshow format.
"I don't think we're done with Oprah Winfrey yet," the Gossip Girl actress told The Insider. "I don't think she's done with us yet [either]."
The star added: "It's just a changing of the guards. She's just reinventing herself again… She's an incredible woman."
This week's two penultimate editions of The Oprah Winfrey Show featured clips from a recent all-star celebration thrown in her honour at Chicago's United Centre. Madonna, Tom Hanks, Tom Cruise and Diane Sawyer were all on hand to pay homage to the 57-year-old TV host.
Winfrey herself described the gala as a night that most people never get to see while they're alive.
The Oprah Winfrey Show wraps up its historic run with an episode reportedly focused on the host's favourite moments from the past.
