Lives will be put in jeopardy and other local residents will be left injured as the flames spread and the entire first floor of the building collapses, The Mirror claims.
Yusef Khan (Ace Bhatti), Masood Ahmed (Nitin Ganatra), Tamwar Masood (Himesh Patel) and Phil Mitchell (Steve McFadden) are expected to be among those inside the B&B during the devastating scenes.
Masood, Tamwar and Phil reportedly end up in hospital with their injuries, while Yusef has long been tipped as a possible fatality.
EastEnders bosses are believed to have filmed the blaze on a special replica set of the B&B, which was built at the Elstree Film Studios.
An EastEnders source told the newspaper: "This is the biggest fire EastEnders has ever seen and there will be casualties. It is such a huge disaster that it will affect most of the families on the show in one way or another."
The insider added: "There is a great tradition at EastEnders of captivating Christmas Day storylines and this year is no different."
