Cleopatra: Portrait Of A Killer is to be based around forensic evidence found in a tomb, including human remains from victims thought to have been killed by the monarch's lover Marc Anthony.
The programme will also examine her marriage to Caesar and the murder of her brother Ptolemy.
BBC specialist factual commissioning editor Mark Bell remarked that the film provides "a fresh perspective" on one of the most famous women in history.
"It may be ancient history, but it still makes a sensational story today, and I'm delighted to be bringing it to BBC One," he said.
