Flack replaces Kirsty Gallagher, who announced she was leaving the Sky1 show in August. Veteran gladiator Wolf, real name Michael Van Wijk, will also be a permanent feature in the revamped entertainment show as a team mentor.
"I'm so excited to be involved in the new series of Gladiators," said Flack. "The show has captured the public's attention all over again with a whole host of new fans, amazing athletes and a healthy dose of Lycra.
"It's great to be able to play a part in something that's struck a chord with so many people over the year. I can't wait to get stuck in."
Flack's presenting credits included the International Pepsi Chart Show, The Games: Live At Trackside, TMI and Big Brother's Big Mouth.
Gladiators returns to Sky1 in January 2009.
