Brydon, who recently released a Comic Relief single as his Gavin And Stacey character Bryn West, will be joined on the show by team captains David Mitchell and Lee Mack.
"I'm a big fan of Would I Lie To You? and I'm really looking forward to joining David and Lee in my new role as host," said Brydon.
It was recently announced that Brydon is to co-host I'm Sorry I Haven't A Clue alongside Stephen Fry and Jack Dee.
