The show, which will air on US network The WB from the autumn, stars Jared Padalecki and Jenson Ackles as Sam and Dean Winchester; two young brothers "bound by tragedy and blood to a dangerous, 'other-worldly' mission."
"This was the stand out show from the United States this season and we are delighted to have secured it for ITV," said Nigel Pickard, ITV's director of programmes.
Supernatural will air on ITV2 or ITV4 - as well as in a late night slot on ITV1 - from early next year.
