Fern, which will air every day, will feature a mixture of celebrities, entertainment and real-life stories.
In each episode, Britton will talk to audience members about their experiences and will offer them advice. The celebrity guests appearing on the programme will also be encouraged to interact with the audience.
Channel 4's head of daytime Helen Warner said: "I have long been a fan of Fern and am thrilled that she is joining us at Channel 4. Whether she's interviewing ordinary people, Hollywood stars or prime ministers, there is no-one better than Fern. For me, she is the British answer to Oprah."
Meanwhile, Britton revealed that she is "very excited" about the project, saying: "I'm working with a great team and together we shall rise to the challenge."
Fern is expected to begin in March.
