Ross, who defected from the BBC last year following the Sachsgate scandal, will front the first edition of his new ITV1 chatshow next month.
The Jonathan Ross Show will be shown straight after The X Factor on Saturday nights.
Sex and the City star Parker and Formula 1 ace Hamilton had not previously been tipped to appear on the programme, although it was rumoured over the weekend that 21 singer Adele would be among the first guest lineup.
Ross's previous BBC One format, Friday Night with Jonathan Ross, regularly averaged between 3.5 and 4m viewers.
The Jonathan Ross Show premieres September 3 at 9.30pm on ITV1.
