The programme - which will air live every day for a week - will focus on a couple who are about to get married.
They bring their extended families together in a country manor for Christmas to see how well they can get along.
The participants will face games, twists, surprises and celebrity guests in an attempt to win up to £500,000.
"This is going to be the perfect show to help build up the excitement of the festive season," Kay said. "It's a whole lot of fun as we try and give away half a million pounds."
Meanwhile, Channel 4's entertainment commissioner Syeda Irtizaali said: "We're excited to broadcast a festive entertainment series that captures the true essence of Christmas for families up and down the country.
"We're also delighted to bring back Vernon to Channel 4. He is the perfect host to bring this together with warmth, sparkle and good humour."
Other Christmas programming on Channel 4 includes special episodes of Coach Trip and Made In Chelsea.
Home for the Holidays will begin on December 18 on Channel 4 and air live every night until Christmas Eve.
