Following on from August's one-off event, House Party: NYE will bring in 2013 with a six-hour live music show featuring a line-up of leading DJs and VJs.
BBC Radio 1's Annie Mac will be one of six international DJs that will transform house parties, bars and living rooms into dancefloors from just before midnight on December 31, 2012.
Channel 4 Arts Commissioner Tabitha Jackson said: "There couldn't be a better night than New Year's Eve for us once again to turn the nation's living room into the ultimate interactive House Party.
"Celebrating with the best artistic talent, Channel 4's House Party: NYE will not only offer an innovative alternative to the usual New Year TV diet but will be the biggest and best party of the year. Pressure's off, New Year's Eve is already sorted."
Annie Mac - who included a set from Jessie Ware during her slot in August's event - will kick off the New Year show, with five further acts to be announced.
BBC Radio 1Xtra's Charlie Sloth will be master of ceremonies for the event, with more special guests to be revealed at a later date.
House Party: NYE will also be an interactive event, with social media being utilised for viewers to become involved with the live broadcast via shoutouts, requests, photos and videos.
