Taking place on Friday, March 21, the charity fundraiser will include a Strictly Come Dancing special featuring four Paralympic stars paired up with professional partners.
Sport Relief Does Strictly will see the judges selecting their favourite partnerships, after the Paralympians and professionals take on a group dance challenge.
Gary Lineker, Gabby Logan, Claudia Winkleman, Jack Whitehall and Clare Balding will be among the presenters on the night.
The star-studded show will also include various celebrity performances, comedy sketches and highlights from this year's Sport Relief challenges.
Davina McCall will take part in the BT Sport Relief Challenge: Davina - Beyond Breaking Point, in which she will travel across the UK by either running, swimming or cycling for seven days.
Sport Relief's Top Dog will be launched in February, featuring celebrity team captains and members of the public in order to find the nation's most talented pooch.
David Beckham will also make a cameo appearance in a one-off Only Fools and Horses sketch on March 21.
On Sunday, March 23, Alex Jones and Matt Baker will host the Sport Relief Games Show from Queen Elizabeth Olympic Park. The programme will show highlights from the weekend's nationwide challenges, which invites members of the public to run, swim or cycle at the Sainsbury's Sport Relief Games.
Little Mix will release a cover of Cameo's 'Word Up' for Sport Relief on March 16.
