The classic sketch, written by the late Ronnie Barker, sees the comedian play a workman attempting to buy "fork handles" from co-star Ronnie Corbett at a hardware store.
It has frequently been named as one of the best sketches of all time and was voted the greatest Two Ronnies skit in a TV special.
The script first emerged on BBC One's The Antiques Roadshow last year and Corbett later confirmed its authenticity.
Peter Spicer of The Original Memorabilia Company, which is handling the online sale, said: "The national interest in this script has been quite phenomenal. It seems that 'Fork Handles' has touched an awful lot of us in some way."
A total of 12 series of the show were made between 1971 and 1987, attracting more than 17 million viewers at its peak. Barker died in 2005 aged 76.
