Mulhern told Digital Spy that he hopes a stand-up will succeed last year's champions Ashleigh and Pudsey as show winners.
"I would love a comedian to win," Mulhern admitted. "It's the hardest thing because they get a short amount of time and you never know what the judges are going to be like.
"If they're in a bad mood, you do get a shorter amount of time - whether you think it's fair or not, that's how it is and that's how it always will be."
The presenter also dismissed rumours that he is planning to quit Britain's Got More Talent due to his commitment to ITV's Catchphrase revival.
"No, that rumour was nonsense," said Mulhern. "I would never want to quit Britain's Got Talent - I really enjoy doing it and all of us are back this year."
Britain's Got Talent returns to ITV on Saturday, April 13 at 7pm.
