The first round of auditions will occur on June 10 and June 11 at Manchester United's stadium Old Trafford in Greater Manchester.
Further auditions are planned for June 18 and June 19 at the Emirates Stadium in London, which is home to Arsenal.
Auditions start at 8.30am and finish at 6pm on all four days and applicants are asked to bring a form of photo identification with them.
Prospective contestants must have a legal right to live in the UK or Ireland and be aged 18 or over by July 31, 2011. They must apply for the show themselves and cannot apply on behalf of another person.
It is not necessary to complete the online application form in order to attend the open auditions, although people who have already applied online will not be barred from attending face-to-face auditions.
Jeff Ford, Channel 5 director of programmes, said: "It is a really exciting time for us as we scour the country to find the next generation of Big Brother housemates. It really couldn't be easier to apply this year - all potential housemates need to do is turn up on the day to one of these two iconic venues, and do everything they can to convince our casting panel that they have what it takes.
"My message to anyone tempted to apply is go for it - in a few months time you might end up a resident in the most famous house in Britain."
Gavin Henderson, creative director of Big Brother, added: "We are on the hunt for housemates that will make this an outrageously fun and unforgettable series of Big Brother. You know where to be and when - we're coming to get you."

Channel 5 bought the rights to the show earlier this year. It will air a series of Celebrity Big Brother this August ahead of the first full non-celebrity run in September.
