According to Entertainment Weekly, the actress will star in one episode of the ABC show.
She was recruited for the role by executive producer Greg Berlanti, who worked with her on Dawson's Creek.
"I went and I begged her and she said, 'I'd love to'," said Berlanti. "[She is] an incredible actress and a good friend."
Holmes will make her guest appearance on the programme in the autumn.
