Rosie's Variety Show will feature celebrity guests, music, comedy skits and prize giveaways.
The hour-long live special may lead to a full series if popular, according to The Hollywood Reporter.
"This is a genre that everybody is anticipating is going to come back," said executive producer David Friedman.
"Rosie sort of invented the daytime variety show. Without question, Rosie is the right person to bring this back."
NBC's Craig Plestis said: "This will reinvent the idea of what a variety show is for 2008. There's very few performers willing to put themselves out there live."
