The Rules Of Attraction star will play a film director looking to adapt a book written by Chad Michael Murray's character to the screen.
For Van Der Beek, the appearance means a return to Wilmington, North Carolina, where he filmed teen drama Dawson's Creek for five years.
One Tree Hill executive producer Mark Schwann commented that he wanted to write a role for Van Der Beek as the series shares similarities with his former show - including the set.
"The spectre of Dawson's Creek is always around us. We're about to complete 128 episodes, which is what Dawson's did ... So I thought it would be cool to get James," he said.
